Laboratory of molecular genetic research


Focus area of the laboratory

— development and further use in the breeding of systems of molecular markers for selecting sunflower genotypes with complex resistance to oil crops pathogens, which will increase the efficiency of breeding in developing lines and hybrids resistant to a complex of pathogens;

— development and further use in the breeding of DNA-marker systems to identify the genes that control the important breeding traits of oil crops, which will increase the efficiency of breeding in developing new genotypes with a given set of economically important traits;

— development of new and optimization of existing systems of DNA markers for molecular-genetic identification and certification of breeding samples, lines, hybrids, and varieties of sunflower, soybean, flax, and rapeseed from the collection of V. S. Pustovoit All-Russian Research Institute of Oil Crops, which will allow studying the molecular genetic diversity of these crops, to estimate the genetic distance between genotypes for selection of parent pairs in hybridization, to certify modern varieties for their further identification, determination of typicality and copyright protection;

— development of molecular genetic passports for new lines and hybrids of sunflower, soybean, flax, and rapeseed varieties of the collection of V. S. Pustovoit All-Russian Research Institute of Oil Crops based on molecular genetic markers.

Laboratory results for 2019-2022

— dominant and codominant molecular genetic markers of mutation of high oleic content were identified and validated for lines of V. S. Pustovoit All-Russian Research Institute of Oil Crops with different content of oleic acid in sunflower seed oil;

— promising DNA markers were found to identify Pl6, Pl8, and Plarg genes controlling the resistance of sunflower to downy mildew pathogen in the collection of lines of V. S. Pustovoit All-Russian Research Institute of Oil Crops;

— we excluded the possibility to locate the gene Or7 controlling resistance of sunflower to race G of broomrape in close proximity to SSR loci ORS 683, ORS 1040, and ORS 1112 on chromosome 3. The region of a further search for the location of the Or7 gene on the linkage map was determined;

— the system of microsatellite markers for genotyping oil flax varieties of breeding of V. S. Pustovoit All-Russian Research Institute of Oil Crops was developed;

— the genetic structure of 4 promising oil flax varieties has been determined using 12 polymorphic microsatellite loci;

— the systems of microsatellite markers for genotyping sunflower and soybean lines and hybrids are optimized, including the use of fragment analysis by capillary electrophoresis;

— about 150 lines, hybrids, and varieties of sunflower, soybean, and oil flax were genotyped by microsatellite markers and the degree of their genetic diversity was determined;

— the work on the development of a microsatellite loci system for genotyping of rapeseed of breeding of V. S. Pustovoit All-Russian Research Institute of Oil Crops has been started. Optimal amplification regimes were determined for the genotyping of rapeseed lines and hybrids using 7 primers for 20 rapeseed lines.
